Recommended operating procedure
The protocol for collagen type I coating on the surface of UpCell (For the case of HUVEC cells).
Materials
- UpCell 3.5cm dish
- HUVEC cells (Lonza, C2517A), medium (Lonza, EGM-2)
- Collagen I (3.34mg/mL) (BD Bioscience, 354136)
- 0.02N Acetic acid solution
- PBS
Collagen coating protocol
We recommend you to increase the concentration of the material by 5 to 10 times to the normal protocol and coat the UpCell surface with collagen solution and incubate at 37 degrees C.
- Prepare 500μg/mL of collagen solution by 0.02 Acetic acid solution.
- Add 1mL of the collagen solution onto the surface of UpCell 3.5cm dish and coat all the entire surface.
- Incubate for 1 hour at 37 degree C incubator.
- Remove the collagen solution from UpCell and wash with pre-warmed 1mL of PBS.
Culture protocol
- Seed HUVEC cells (10,000 cells/cm2) onto collagen coated UpCell 3.5cm dish.
- Incubate at 37 degree C incubator for over 2 days until cells overconfluence.
