Recommended operating procedure
Standard Operating Protocol (SOP) for collection of cell-sheet.
- Culture cells beyond over-confluency. Please ensure that cells are covering entire surface of the cultureware.
- Take out UpCell from the incubator. Remove most but not all of the culture media from UpCell to prevent cells from drying.
- Leave UpCell at room temperature (20-25℃) for 20-30 minutes. (*1)
- Scrape around the edge of the dish to cut off the cells that may be attached to the corner of UpCell. Please confirm that edge of the cells are starting to detach before moving on to next step.
- Place membrane on top of the cell-sheet. Make sure there is no air bubble in between cell-sheet and the membrane. (*2)
- Gently peel membrane off from the surface using forceps. Make sure that cell-sheet is firmly attached to the membrane.
*1 Placing membrane before temperature reduction (but always after removal of culture media) will also work.
*2 Please note that membrane can easily round up when soaked initially in the culture media. Resist round up by using forceps. Apply gentle pressure to the membrane where it started to round up for 20-30 seconds.
