Can you separate molecules that are insoluble in water?
Yes, you can. For example, you can separate methanol which is affinity with water by dissolving it with THF.
Can you use organic solvents as an eluent?
You can still use organic solvents. However the real value of "Aqua Way" lies in the fact that it does not require organic solvents to carry out separation/analysis. The core values it provides are cost reduction, concept being consistent with ecology, maintains the activities of the molecules and so on.
Can you shorten the separation time?
You can shorten the separation time by gradienting the temperature. You can obtain the same consequence as if you are using organic solvents by gradienting the temperature from high to low.
What is the suitable temperature range for this column?
Analysis is usually done in the range of 5°C~50°C. The retention characteristics suddenly changes at PIPAAm's LCST of 32°C. Please adjust the temperature to suit your target molecules. Our "Temperature gradienter" allows precise setting and control of the temperature. It can be easily operated with your PC.
I see the peak overlap each other. What should I do?
Add some NaCl to the buffer. The LCST of PIPAAm can be changed freely by adding NaCl to the eluent. By adding NaCl to the eluent, the LCST is lowered and you can separate the molecules at lower temperature.
Are there anything to note when using H2O or buffer as an eluent?
When using buffer as an eluent, please make sure to replace the whole system with same buffer. Otherwise reproducibility may be lost.
I don't get the sharp peak by using H2O as an eluent. What is the problem?
Change the eluent from H2O to buffer. The characteristics of the target molecules may be influenced by such variables as pH. A sharper peak can be obtained by using suitable buffer adjusted for target molecules.
The pressure is gradually increasing as I continue to use. What should I do?
There may be some foreign substance in the column.Cleanse column with cold water. It is always advisable to use filter when injecting the sample.
How to cleanse the column?
Cleanse column with cold water (about 5°C). The surface of the beads becomes hydrophilic at 5°C therefore you can easily wash away hydrophobic substance such as proteins.
What should I do when not using column for a time being?
First of all, please cleanse the column thoroughly with cold water. If not in use for a long period, replace eluent with H2O/MeTH(50/50(v/v)). (The eluent will rot if left with 100% of H2O for a long period.)
Is it suitable for quantitative analysis?
It can be said that "Aqua Way" possess a remarkable advantages for quantitative analysis. Since it does not use organic solvents as a mobile phase, you can reduce the background noise when using UV or fluorescent detector. You can obtain stable base line even if lower band of wavelength is tuned in. Besides, time required till the system stabilizes is very short.
What kind of detector can you use?
You can use such detecotr as UV, fluorescent and LC/MS.
